What is chondroitin sulfate?

Chondroitin sulfate, a natural glycosaminoglycan, is found in cartilage and the extracellular matrix. Plays a key role, among others. in the regulation of cell development, proliferation (ability to reproduce) and cell differentiation (1). Chondroitin sulfate (in addition to glucosamine sulfate) showed a beneficial effect on the metabolism of cell models derived from synovial joints: chondrocytes, synoviocytes (cells of the synovial membrane of the articular capsule) and subchondral bone cells. It increased the synthesis of collagen and type II proteoglycans in human articular chondrocytes and influenced the anabolic-catabolic balance of the cartilage extracellular matrix (2). What else do we know about chondroitin? Well, it is the main component of the extracellular matrix of the articular cartilage, which is important in creating a significant osmotic pressure. Thus, it can provide the cartilage with strength and flexibility to withstand tensile stress under load conditions (3).

 

Why is hyaluronic acid important?

Hyaluronic acid is a polysaccharide belonging to the glycosaminoglycans. It is a component of connective, epithelial and nervous tissue and is an important component of the extracellular matrix. The average molecular weight of hyaluronic acid may influence its physicochemical properties. Among the many biological effects, hyaluronic acid takes part, for example, in cell differentiation and viscoelasticity (4). The hyaluronic acid molecule is especially present in the synovial fluid with excellent viscoelasticity, high moisture retention capacity, high biocompatibility and hygroscopic properties, thanks to which it acts as a lubricant, shock absorber, stabilizer of the joint structure and regulator of water balance and flow resistance (3).

What connects proline with collagen?

Proline comprises about 10% of all amino acids in collagen, which is one-third of mammalian proteins. Collagen, as the most abundant protein in the body, is essential for maintaining the normal structure and strength of connective tissue such as bones, skin, cartilage and blood vessels. Proline support is required for the biosynthesis of collagen as well as other proline-containing proteins. Therefore, the regulation of proline availability for collagen biosynthesis is of key importance for maintaining tissue integrity (5).

 

Effect of vitamin C on collagen

Vitamin C is a water-soluble molecule with significant antioxidant properties. By acting as a powerful antioxidant, it neutralizes harmful reactive oxygen species. It helps to protect cells against oxidative stress. Vitamin C plays a role in collagen synthesis - it is necessary for the formation of collagen fibres and cell differentiation. Vitamin C supports the proper production of collagen to ensure the proper functioning of blood vessels, bones, cartilage, gums, skin and teeth (6, 7).
