Collagen is the most abundant protein in the human body, where it is responsible for structure, stability and strength, especially in the skin layers. Already in the mid-1930s, it was shown that collagen consists of a regular structure at the molecular level. With regard to ageing, collagen (and elastin) deposition decreases with chronological ageing (over time), and especially with photoaging (sun exposure). In addition, it can be broken down by protein hydrolysis. Collagen supports various tissues such as tendons, ligaments, skin, teeth, and many other connective tissue structures. Collagen is a viscoelastic material with high tensile strength and low immunogenicity (i.e. it has a poor ability to induce a specific immune response against itself) (1).
Collagen has a double effect in the skin, where it first supplies the building blocks of collagen (and elastin), and secondly, it binds receptors in fibroblasts located in the layers of the dermis, stimulating the synthesis of collagen, elastin and hyaluronic acid (1).
Hyaluronic acid is a component of the body that is present in every connective tissue and organ, including such as skin, synovial fluid, blood vessels, serum, brain, cartilage, and heart valves. In particular, the skin has the highest amount of hyaluronic acid in the body, with 50% of the body's total hyaluronic acid being present in the skin (2). Regarding the functions of hyaluronic acid, it is an essential component of the extracellular matrix of the skin and plays an essential role in the metabolism of the dermis. It is one of the most hydrophilic molecules found in nature and can be described as a "natural moisturizer" (3).
Unfortunately, that's the way it is. Under the influence of various factors (e.g. ageing, ultraviolet light, dryness, etc.), the hyaluronic acid content of the skin decreases, as does the water content of the stratum corneum, which leads to wrinkles and ageing of the skin. Research shows that the amount of hyaluronic acid contained in the skin gradually decreases with age. More precisely, its content in the skin of people aged 75 years is less than a quarter of the content of hyaluronic acid in the skin of people aged 19 years (4).
Vitamin C helps in the proper production of collagen to ensure the proper functioning of blood vessels, bones, skin, cartilage, gums and teeth (5). This is because vitamin C acts as a cofactor for proline and lysine hydroxylases, which stabilize the tertiary structure of the collagen molecule and also promote the expression of collagen genes. In addition to stabilizing the collagen molecule by hydroxylation, vitamin C also stimulates the production of collagen mRNA by fibroblasts (6).
